Pseudomonas putida Tryptophan Synthetase : Partial Sequence of the a Subunit IRVING

چکیده

There is 50% identity in the sequences of the first 50 residues of the a chains of Escherichia coli and Pseudomonas putida. No deletions or additions of residues are found in this region, except for the N-terminal methionine residue which is missing in the polypeptide isolated from P. putida. Most of the residues which differ are chemically dissimilar, and half of them are specified by codons which differ by more than a single base. The two residues known by mutational analysis to be essential for catalysis in E. coli are conserved in P. putida. The potential taxonomic usefulness of information of this sort is analyzed.

Amino acid sequences of fifty residues from the amino termini of the tryptophan synthetase chains of several enterobacteria.

The sequences of the first 50 residues of the tryptophan synthetase o( chains of Shigella dysenteriae, Salmonella typhimurium, and Aerobacter aerogenes were determined and compared with the corresponding sequences of Escherichia coli and Pseudomonas putida. On the basis of observed amino acid residue differences, and inferred nucleotide differences in the respective genes, similarity to the E. ...

Up-promoter mutations in the trpBA operon of Pseudomonas aeruginosa.

In Pseudomonas aeruginosa, the operon encoding tryptophan synthase (trpBA) is positively regulated by the TrpI protein and an intermediate in tryptophan biosynthesis, indoleglycerol phosphate (InGP). A gene fusion in which the trpBA promoter directs expression of the Pseudomonas putida xylE gene was constructed. By using a P. putida F1 todE mutant carrying this fusion on a plasmid, three cis-ac...

Transcriptome analysis of a phenol-producing Pseudomonas putida S12 construct: genetic and physiological basis for improved production.

The unknown genetic basis for improved phenol production by a recombinant Pseudomonas putida S12 derivative bearing the tpl (tyrosine-phenol lyase) gene was investigated via comparative transcriptomics, nucleotide sequence analysis, and targeted gene disruption. We show upregulation of tyrosine biosynthetic genes and possibly decreased biosynthesis of tryptophan caused by a mutation in the trpE...

Comparative genomics of an endophytic Pseudomonas putida isolated from mango orchard

We analyzed the genome sequence of an endophytic bacterial strain Pseudomonas putida TJI51 isolated from mango bark tissues. Next generation DNA sequencing and short read de novo assembly generated the 5,805,096 bp draft genome of P. putida TJI51. Out of 6,036 protein coding genes in P. putida TJI51 sequences, 4,367 (72%) were annotated with functional specifications, while the remaining encode...

Structure and regulation of the anthranilate synthase genes in Pseudomonas aeruginosa: I. Sequence of trpG encoding the glutamine amidotransferase subunit.

We have determined the DNA sequence of the distal 148 codons of trpE and all of trpG in Pseudomonas aeruginosa. These genes encode, respectively, the large and small (glutamine amidotransferase) subunits of anthranilate synthase, the first enzyme in the tryptophan synthetic pathway. The sequenced region of trpE is homologous with the distal portion of E. coli and Bacillus subtilis trpE, whereas...
